Although Glaxo is still years away from having an FDA approved drug for sale, Pfizer, a major Glaxo competitor, has begun the competitive battle, casting doubt on Glaxo’s claims.  Yet Pfizer’s own researcher, Dr. Kay Ahn was quoted in an article last month in Nature magazine, and downplayed the significance of her study.  “Under our conditions we didn’t see beneficial effects, but we don’t want to make a big conclusion out of those results,” she said.

Glaxo said in a statement that “scientific debate around the interpretation of data is a normal part of discovery in emerging areas of research.” It maintains that Pfizer’s interpretation of data “is at odds with multiple studies conducted at Sirtris and those published by independent investigators.” The company added that the “purity of the compounds that were used in their study … cannot be ascertained due to the lack of data provided. The publication also does not cite nor discuss multiple scientific publications which are in direct contrast to their primary conclusions; as such it is unclear how to place this work in perspective of what has been found by other investigators.”

New Miller School Mitochondria Research Could Impact Effects of Aging

A research group led by Carlos T. Moraes, Ph.D., professor of neurology and cell biology and anatomy at the Miller School, has published new findings about how increased mitochondrial levels may improve overall health and increase longevity.

According to the article published in the Proceedings of the National Academy of Science, the researchers increased the amount of mitochondria in a mouse model by a transgenic expression of a gene called PGC-1α. The protein produced by this gene controls the expression of many genes that are involved in mitochondrial biogenesis.

There are numerous human clinical trials, and studies by the National Institute of Environmental Health, and even animal studies to prove it. Of course, companies that sell a low-dosage  ( 50 – 200 mg of Resveratrol) — will try to convince you otherwise. Those companies will tell you that you either don’t need more quantity. They may even tell you that their resveratrol is “more potent”. This just doesn’t make sense.

In 2002, the National Institute of Environmental Health Sciences commissioned a report entitled, “Trans-resveratrol Review of Toxicological Literature.”  A review of more than 200 referenced scientific studies were included, and the report stated that there were no reported adverse effects from Resveratrol on humans.

Animal studies have used the human-equivalent of over 1,000 mg (10 mg/kg of bodyweight) of Resveratrol, and have shown no toxic effects. Massive dosage studies were also performed on animals. “The no observed adverse affect level (NOAEL) was 300 mg/kg/day,” which would be a human equivalent dosage of 30,000 mg or 10 – 100 times the maximum concentrations contained in human high-concentration oral nutraceutical dosages, such as in Vinomis’ Vindure 900 and others.

Human studies also confirm the safety and effectiveness of Resveratrol in highly concentrated forms. Studies including those at Marywood University, University of Leicester (UK) and University of Michigan, etc. all found no significant side effects and no evidence of toxicity

In 2006, Xenomis conducted a study on humans with Dr. James Smoliga of Marywood University (2006) using the formulation:

“I beleve there are one or two articles examining the pharmacokinetics of it [dosage] (combined with safety) which found it to be safe in up to 5g [5,000 mg] doses, I believe.  It appears that with dosages in the 400 mg range, it reaches a peak and then subsides, whereas in the 1g (1,000 mg) or more range, higher blood concentrations are maintained.  However, I believe it is not yet certain whether which would be more advantageous – to have a rise in fall in resveratrol concentration or a steady level of resveratrol.  Based on some of the physiology, I would think a rise and fall mechanism would actuallly be more beneficial, and if this is the case the 400mg dosage should work quite nicely.”

“We used ~5-30 mg/kg in our mice, and Johan Auwerx used 200-400 mg/kg.  If you subscribe to the “allometric scaling” by body surface area method, you divide by 12 to go from mouse to human, yielding ~150 mg or ~2g [2,000 mg] as the respective human-equivalent doses.  But since other polyphenols may have an additive or synergistic effect, I think it’s quite likely that Vindure is delivering the effect of a 2g [2,000 mg] dose of pure resveratrol.”
